A Rapid Micro Method for Determ nation of Ascorbic Acid in Urine by Ferric Reduction

Leon S. Vann 1

1 Cancer Biochemistry Laboratory, Department of Public Health, State of California, Berkeley, Calif. 94704.

A new method for the determination of ascorbic acid in urine is based on the quantitative reduction of ferric to ferrous ion by ascorbic acid and the colorimetric measurement of the ferrous ion through its formation of a colored complex with bathophenanthroline. The complex is quantitatively extracted into chloroform, and its absorbance is measured in this solvent. The method is simple, very rapid, and has good specificity for clinical purposes. Only 0.05-0.25 ml. of urine is required per determination. All reagents used in the method, except the ascorbic acid standard, are stable indefinitely.
