A gas chromatographic method for the estimation of microgram quantities of hydroxy-amobarbital in urine is described. The method has been applied to a study of the metabolic fate of therapeutic doses of amobarbital (60 and 180 mg.) in man. We have found that 45% of an ingested dose of the drug is excreted in urine as hydroxy-amobarbital in 3 days and that small amounts of the metabolite are detectable for as long as 5 days following the ingestion of a single 180-mg. dose of amobarbital.
