Oxidation of urinary pregnane-17,20-diols (17-oxogenic steroids) by sodium metaperiodate is complete within 10 min at 55°C if the concentration of sodium metaperiodate at the end of the reaction exceeds 1.5 x 10-3M. A safe excess of sodium metaperiodate is present if assay samples, after incubation for 10 min at 55°C in 3 x 10-3M ethylene glycol, give a positive test with starch-iodide paper.
