Dear Editor: We read with great interest the paper “Proteomics and Bioinformatics Approaches for Identification of Serum Biomarkers to Detect Brease Cancer”. The authors screened for potential tumor biomarkers in 169 serum samples. Diluted serum samples were applied to immobilized metal affinity capture Ciphergen ProteinChip® Arrays previously activated with Ni2+. Three peaks, designated BC1 (4.3 kDa), BC2 (8.1 kDa), and BC3 (8.9 kDa), were finally selected as potential biomarkers for detection of breast cancer. We performed the proteinchip work to detect breast cancer according to this paper in the serum of 15 breast cancer patient and 15 normals. The conditions we used are the same as mentioned in this article. But contrary to the findings in the paper, mass spectrometry of IMAC-Ni chip did not detect differences in the peaks of BC1(4.3KD), BC2(8.1KD),BC3(8.9KD) between the cancer patients and normals. We were surprised by the opposite conclusion presented in our study in comparison with the published work. We find no difference in the molecular weights of these three markers. We hope the authors can provide further detailed explanations. We also wonder why, given that mass spectrometry can provide extremely accurate molecular weight, the authors showed only the approximate MW.
